GenEvolutioN at EUROTOX 2025: advancing non-animal genotoxicity testing for cosmetics

New approach methodologies: from ethics to scientific relevance

Driven by the New Approach Methodologies (NAMs) movement, GenEvolutioN leverages human-relevant in vitro models to replace animal testing. The RSMN assay, using the 3D reconstructed EpiDerm™ skin model, offers a physiologically relevant system that mimics human skin, accounting for dermal absorption, metabolism, and cell interactions.

This approach enhances the predictive value of genotoxicity testing, particularly when standard 2D assays produce ambiguous or false-positive results.

Scientifically validated and regulatory-aligned

In early 2025, the European Scientific Advisory Committee (ESAC) validated the RSMN assay as a reliable method for assessing skin-applied substances. This milestone supports the growing integration of non-animal approaches under frameworks such as the EU Cosmetics Regulation, which mandates alternative testing methods.

Looking ahead: skin organoids and the future of NAMs

GenEvolutioN is also pioneering the use of skin organoid models, incorporating multiple cell types—epidermis, dermis, adipocytes, neurons, and hair follicles. These advanced systems allow for individualized genotoxicity assessments and open new avenues for safety profiling of innovative ingredients.
